An honest word on the evidence

MOTS-c has the most interesting biology in this group and, like the rest of it, very little human evidence. The work sits mostly in cells and animals; the FDA's advisory committee considered it in July 2026 in the context of obesity and osteoporosis, and recommended it for the 503A Bulks List on a narrow margin against FDA staff's own recommendation. It is often described as an exercise mimetic, which is a phrase from the laboratory rather than a claim anyone has demonstrated in people, and it is not a reason to train less. These peptides sit in an unsettled regulatory position. In April 2026 the FDA removed twelve peptide substances from Category 2 of its interim 503A bulk substances list, this one among them — a step that followed the nominators withdrawing their nominations rather than any safety finding, and one that does not by itself authorize compounding. In July 2026 the Pharmacy Compounding Advisory Committee voted on whether seven of them should be added to the 503A Bulks List. That vote is a recommendation, not a decision: formal addition requires notice-and-comment rulemaking, which has not completed. Nothing here is FDA-approved for any use.
